ODA Chief Executive David Higgins and London 2012 organising committee chairman Sebastian CoeToday's announcement marks the first time Games-time operations and legacy usage have been fully integrated from the outset.
The new masterplan optimises the layout of venues within the park and includes the relocation of the Volleyball competition to Earls Court.
ODA Chief Executive David Higgins said: "Detailed planning now is worth its weight in gold further down the track. This is a key building block for the project that allows us to move to the next stage of development.
"By making such significant decisions so early we also reduce risk and therefore enable cost minimisation."
London 2012 organising committee chairman Sebastian Coe said: "We are delighted that we are setting new standards and benchmarks for delivering the IOC's vision of a sustainable legacy.
"This is the first time a city has both Games effectiveness and legacy enshrined together, less than one year since Singapore and six years to go to the Games."
The Olympic Park masterplan has been approved by the IOC and the relevant sports governing bodies and federations.
